*转录具有选择性和不对称性，一条链为RNA的合成提供模板，称为&#039;&#039;&#039;模板链&#039;&#039;&#039;(又称无意义链或Waston链)。另一条链与RNA编码信息一致，称为&#039;&#039;&#039;编码链&#039;&#039;&#039;（又称有意义链和Crick链）&lt;br /&gt;
*以四种NTP为原料，还需要镁离子（一个镁离子同NTP进入活性中心，进行静电屏蔽）&lt;br /&gt;
*转录不需要引物&lt;br /&gt;
*转录方向为5&#039;→3&#039;&lt;br /&gt;
*转录的忠实性低于复制，但在特定因子作用下可以发挥校对功能（RNS聚合酶通常无3&#039;外切酶活性）&lt;br /&gt;
*有高度的进行性&lt;br /&gt;
*以上为DNA转录的概述&lt;br /&gt;
=RNA聚合酶（RNAP）=&lt;br /&gt;
RNA聚合酶是负责在生物体内催化RNA合成的酶，具体信息如下：&lt;br /&gt;

==依赖于DNA的RNA聚合酶==&lt;br /&gt;
===细菌的RNA聚合酶===&lt;br /&gt;
细菌只有一种RNA聚合酶。&lt;br /&gt;
*组成：由&#039;&#039;&#039;核心酶&#039;&#039;&#039;和&#039;&#039;&#039;σ因子&#039;&#039;&#039;组成全酶。核心酶由2个α亚基、1个β亚基、一个β＇亚基和一个ω亚基组成。σ因子有种属特异性和条件特异性，以大肠杆菌为例，σ&amp;lt;sup&amp;gt;70&amp;lt;/sup&amp;gt;因子起始大多数转录物的转录，用于大多数启动子的识别，但在特殊情况（如热激情况下，σ&amp;lt;sup&amp;gt;24&amp;lt;/sup&amp;gt;,σ&amp;lt;sup&amp;gt;32&amp;lt;/sup&amp;gt;起始相关基因转录）由其他σ因子起始转录。（σ&amp;lt;sup&amp;gt;28&amp;lt;/sup&amp;gt; 专门负责鞭毛相关的mRNA的合成）&lt;br /&gt;
*功能：α亚基的N端参与酶组装，C端参与和调节蛋白与增强元件的作用；两种β亚基共同构成催化中心，其中β’亚基带正电，与DNA结合；ω亚基促进酶的组装并维持其稳定。&lt;br /&gt;
*抑制剂：&#039;&#039;&#039;利福平（利福霉素）&#039;&#039;&#039;作用于β亚基，抑制起始；&#039;&#039;&#039;利（迪）链霉素&#039;&#039;&#039;作用于β亚基，抑制延伸；肝素作用于β＇亚基（抵消正电），抑制酶活性。&lt;br /&gt;
===真核的RNA聚合酶===&lt;br /&gt;
====分类====&lt;br /&gt;
真核生物的RNA聚合酶可分为三类，分别为&#039;&#039;&#039;RNA聚合酶I&#039;&#039;&#039;、&#039;&#039;&#039;RNA聚合酶II&#039;&#039;&#039;和&#039;&#039;&#039;RNA聚合酶III&#039;&#039;&#039;，另外还有&#039;&#039;&#039;细胞器RNA聚合酶&#039;&#039;&#039;。&lt;br /&gt;
&lt;br /&gt;
* 组成：有2个大亚基，加上12~15个小亚基，2个大亚基的一级结构与细菌的两种β亚基相似，含有细菌α亚基的同源物。&lt;br /&gt;
&lt;br /&gt;
*RNA聚合酶I：位于核仁，起始5.8SrRNA、28SrRNA、18SrRNA（以45SrRNA的形式转录），对α-鹅膏蕈碱不敏感，对放线菌素D高度敏感。&lt;br /&gt;
**主要功能：亚基A190相当于β′亚基，亚基A135相当于β亚基，亚基AC40、AC19相当于α亚基，亚基Rbp6相当于ω亚基&lt;br /&gt;
*RNA聚合酶II：位于核质，起始mRNA，lncRNA，多数miRNA，具有帽子结构的snRNA与snoRNA的转录，对α-鹅膏蕈碱最敏感，对放线菌素D轻度敏感。&lt;br /&gt;
**主要功能：亚基Rbp1相当于β′亚基，亚基Rbp2相当于β亚基，亚基Rbp3、11相当于α亚基，亚基Rbp6相当于ω亚基&lt;br /&gt;
*RNA聚合酶III：位于核质，起始tRNA，5SrRNA,7SLRNA，7SKRNA，无帽子的snRNA和snoRNA，端粒酶RNA和多种小RNA转录，对α-鹅膏蕈碱中度敏感，对放线菌素D轻度敏感。&lt;br /&gt;
**主要功能：亚基C160相当于β′亚基，亚基C128相当于β亚基，亚基AC40、AC19相当于α亚基，亚基Rbp6相当于ω亚基&lt;br /&gt;
*线粒体RNA聚合酶：单体酶，与T7噬菌体同源，由核基因编码，负责mt内全部RNA的合成。&lt;br /&gt;
*叶绿体RNA聚合酶：分两种：一种是由核基因编码的单体酶，负责管家基因（包括rRNA）的转录；一种是由cp基因编码的多聚酶，负责光合链上cp自身合成的亚基基因的转录。&lt;br /&gt;
细胞器RNA聚合酶对α-鹅膏蕈碱不敏感，对放线菌素D敏感。&lt;br /&gt;
&lt;br /&gt;
* 所有真核的RNAPII在最大亚基C端，都重复的富含羟基氨基酸残基，构成了特别的羧基端结构域（CTD），是活性必需的。一旦CTD磷酸化，转录就从起始进入延伸阶段。&lt;br /&gt;
&lt;br /&gt;
*此外，植物细胞还有两种特化的RNA聚合酶II（缺乏CTD），为RNA聚合酶IV和V，负责转录lncRNA，介导RNA引导的DNA甲基化。&lt;br /&gt;
====抑制剂====&lt;br /&gt;
*α-鹅膏蕈碱：见上；&lt;br /&gt;
*放线菌素D：见上，作用机制为插入DNA双螺旋中的GC碱基对，阻止聚合酶移动。&lt;br /&gt;
*利福霉素与利链霉素只对线粒体与叶绿体有抑制作用，效果与其抑制细菌时相同。&lt;br /&gt;
===古菌的RNA聚合酶===&lt;br /&gt;
*类似真核生物的特征：不受利福霉素与利链霉素的抑制，不能直接识别启动子，需要转录因子，结构上类似于RNApolII。&lt;br /&gt;
*类似原核生物的特征：只有一种。&lt;br /&gt;
*特殊处：最大的亚基在C端无CTD结构。&lt;br /&gt;
